Responsibilities - Support the Downstream Process Development (purification) group of the Vaccine Production - Program (VPP) at the Vaccine Research Center. - Lead development of mRNA drug substance process, including purification of pDNA,generation and purification mRNA constructs and their subsequent LNP encapsulation. - Collaborate with formulations team to enable identification and selection of appropriate lipids and LNP composition for downstream development activities. - Plan and execute design-of-experiments (DOE) studies & high-throughput screening studies for purification process improvements and advancements including parameter optimization, scale-up, process operating window determination and process capability evaluation. - Lead efforts within the purification group to design, develop and optimize cell culture clarification (depth filtration and/or continuous centrifugation), chromatography, Tangential Flow Filtration (TFF), and Normal Flow Filtration (NFF) processes for all clinical trial vaccine candidates, including antibodies and subunit or nanoparticle vaccines. - Oversee execution of viral clearance validation studies to support IND-submission and collaborate with external stake holders (VCMP/Vendors/CROs/CDMOs) on study logistics. - Perform and support purification of research-phase recombinant proteins, mRNA vaccines, virus vaccines and/or virus-like particles in support of other groups at the VRC. - Interface with key stakeholders (internal and external) to define development strategies and key milestones. - Clearly communicate results and recommendations to collaborators, colleagues, managers, and senior management, leading project decision-making. - Present findings at internal department level to institution level meetings and seminars. - Author and/or review technology transfer documents in support of GMP manufacturing. - Author and/or review relevant sections of IND submissions. - Contribute to research publications through internal and external collaborations. - Pursue novel technologies to advance the capabilities of the VPP. - Review and publish technical reports and method protocols. - Write and review process batch records for toxicology batch productions. Experience Required / Requirements - The ideal candidate will possess a Ph.D. in a Biochemistry, Chemical Engineering, or other life sciences and possess the following: - 3 – 5 years of experience in industry-related downstream development lab. - Prior mentorship experience with proven leadership abilities in team-oriented environment. - Ability to navigate ambiguity and synthesize actionable next steps to move programs and strategies forward. - Experience with mRNA IVT and purification. - Proficiency in commonly used techniques and tools for biologics purification and characterization, including column chromatography, TFF, NFF, UV-Vis spectroscopy, etc. - Demonstrated knowledge of maintaining accurate and detailed records. - Strong organization, communication, and interpersonal skills, with the ability to drive projects in a highly collaborative work environment. - Good understanding/knowledge of the overall drug development process, cGMP, and regulatory requirements. - Familiarity with automation technologies and high-throughput techniques relevant to biologics production and process analyses. - Experience writing technology transfer documents and IND submission sections related to downstream processes. - Knowledge of the process scale-down and scale-up. We are seeking a Sr. Budget Analyst to join the team at Axle Informatics. The selected candidate will be responding to NIH budgetary requests, preparing reports, forecasts, and analyses for NIH customers as well tracking contract metrics including Earned Value Management. Overview Axle is seeking a highly ambitious, adaptable, structured, and detail-oriented Biologist   to join our vibrant team at the National Institutes of Health (NIH), National Institute of Allergy and Infectious Diseases (NIAID),Vaccine Research Center (VRC), Pandemic Repository through Microbial and Immune Surveillance and Epidemiology (PREMISE).  Responsibilities - Perform a wide range of cell biology techniques including cell culture, cloning, binding, and cell-based assays using high-throughput modalities and flow cytometry. - Perform a wide range of molecular biology techniques including qPCR, DNA/RNA analysis, - DNA/RNA isolation and purification, next generation (Illumina) sequencing, transfection, and gene cloning. - Conduct research including planning and conducting experiments and data analysis. - Integrate research findings into existing bioinformatics workflows. - Develop, troubleshoot, and adjust scientific methodology to correct problems and deficiencies in a research project or experiment. - Maintain accurate, daily records of experiments. - Organize daily workload based on priorities, rotation schedule and needs of the laboratory. - Use a wide variety of laboratory information systems, statistical methods, and computer programs to collect, organize, and analyze data for reporting of verified results. - Independently analyze and discuss the interpretation, reliability, and validity of test data. - Communicate and consult effectively with PREMISE and laboratory staff. Curriculum design activity from these projects will require expert-level pharmacoepidemiologists to evaluate, curate and prepare content. Publication opportunities may arise from the curriculum development focus.   The position will be based at Axle in Bethesda, Maryland, but functionally remote from US locations, with occasional travel. The successful applicant will be involved in designing and producing RWE curriculum using N3C observational studies in support of the National Center for the Advancement of Translational Science (NCATS).   The ideal candidate will have experience in the application of causal inference methods (IPTW, PSM, G-methods, etc.) and observational study designs (target trial emulation, self-controlled case series, test-negative design, etc.) They will have experience leading RWE projects from start to finish and as author for publications in the field. The candidate will have a deep understanding of the limitations of Real World Data (RWD) and various forms of bias and confounding that arise in observational studies (immortal time bias, confounding by indication, time-varying confounding, etc…). They will work with clinicians, biostatisticians, and other stakeholders to identify educational curriculum of significant public health importance. They will stay current with scientific and technical literature and remain informed about new approaches and methodologies, including participation in conferences or workshops, independently evaluating scientific resources, and potentially publishing scientific papers from this work.   Responsibilities - Develop RWE curriculum for AIM-AHEAD extension of N3C. - Collaborate with instructional designers to create new epidemiology courseware. - Serve as a RWE subject matter expert to Axle and NCATS leadership. - Implement recommendations from leadership on standardization, reproducibility, and data transparency. - Develop standards for the operationalization and fit-for-purpose use of Causal and AI/ML methods applied to Real-World Data (RWD). - Produce criteria to evaluate the incremental benefit of PPRL-linked datasets (i.e., improvements in the quality and/or scope of analyses, reduction in data missingness, etc.). - Participate in code and documentation peer review, integration, unit testing, and validation. - Produce exemplary material relevant to use of RWD including but not limited to treatment of missing data, case control selection, minimization of type I/II error, bias, prevention of model overfitting and poor generalization and approaches to Electronic Health Record data limitations. - Participate in preparing reports, visualizations, proposals and publications. - Present your work internally at Axle, to the NIH and at selected conferences. - Support investigations requiring biostatistical and/or machine learning knowledge and domain expertise. Responsibilities   - Gain and maintain a comprehensive understanding to ACTIV3 protocol and, as requested, other COVID-19 protocols - Identify and obtain resources needed to establish and maintain DCR’s International Coordinating Center as high-functioning and capable of meeting and exceeding the INSIGHT requirements for its ICCs - Provide senior level oversight of the work of the DCR ICC team including setting deadlines. Identify and resolve barriers to task completion. - Provide assistance and guidance to executive level management - Provide recommendations to DCR leadership for decisions / actions related to the DCR ICC - Assist with the development and implementation of ICC operational framework including processes, - leveraging existing INSIGHT ICC procedures and processes whenever possible and appropriate. - Assist with the development and implementation of the strategy for coordination of clinical trial - operations in concordance with NIAID DCR leadership. - Ensure adequate resources are available for the following areas of trial management: - Safety - Monitoring - Training - Pharmacy - Laboratory - Human subjects protection - Regulatory adherence - Communications - Support sites that are conducting or are interested in conducting the trial(s) in identifying and - resolving gaps in staffing. - Review enrollment and determine areas of need - Develop strategies to increase enrollment - Develop strategies to optimize site participation and adherence to protocols - Liaise with other agencies and organizations when necessary - Liaise with consultant groups to optimize performance - Support DCR in identifying potential sites for participation in the trial(s) - Participate in engagement with, and addition of, new sites - Develop and implement standard operation procedures for trial implementation - Participate in clinical trial agreement negotiations and discussions with sites and networks - Participate in the Quality Assurance/Quality Control processes - Prepare reports for DCR leadership as requested - Assist with developing budgets and monitoring costs Experience Required / Requirements - Masters degree in related field or equivalent experience - Minimum of twenty (20) years’ experience in related field - Expertise in management and operations - Expertise in the fundamentals of clinical trials - Experience with IRB and FDA requirements - Experience in clinical research setting with protocols and clinical trials - Excellent writing skills - The position will involve frequent interaction between NIAID Federal and other contract staff,and sites interested in, or engaged in ACTIV3 or other NIAID COVID-19 clinical trials. Demonstrate scientific rigor in assessing own data and that of others. - Effectively present results at internal meetings. - Responsible for meeting project goals within time and budget constraints, including vaccine projects, but not limited to vaccines. - Demonstrate ability for scientific insight and conceptual thinking. Specifically, should be able to clearly define problem statement and make scientific hypothesis. - Independently plan, design, and execute projects within a specific scientific field (bio-separation technology) utilizing good scientific judgment. - Typically conduct more than one vaccine project at a time. - Develop research findings and makes recommendations. - Utilizes forward-looking perspective to contribute to “go/no go” decisions. - Make recommendations on outside technologies for company to pursue.   Experience Required / Requirements - Ph.D. in a scientific field. Three to five (3+, 5+) years of specialized experience plus a Master’s degree is equivalent to a Ph.D. Five to eight (5+, 8+) years of specialized experience plus a - BA/BS degree is equivalent to a Ph.D. Minimum of nine (9) years of experience in a science related field is required. - Demonstrates expert knowledge working in the research laboratory environment. Computer skills to include MS Office Suite. - Must be a team player who can effectively work with members from cross-functional departments. - Demonstrated knowledge of maintaining accurate and detailed records. - Strong oral and written communication skills. - Familiarity with computer software including word processing, data evaluation, and project timeline management. - This position is for bio-separation scientist. Experience is preferred on capillary experience, chromatographic analysis of protein molecules.
